The practice will be located at 1 West Auckland Road, Cockerton District Centre, in a site previously used as a beauty salon.
Darlington Borough Council approved the application, submitted by Mr Sunil Kumar, on Thursday which will see the former beauty salon changed to a private GP practice.
It will be open Monday to Saturday from 8.30am to 9pm, and Sundays and bank holidays from 8am to 6pm.
The new practice is expected to double current employment, increasing from 1.5 full-time equivalent roles to three, with two full-time and two part-time staff members.
Accessibility improvements have been a key part of the plan, with a level pedestrian access ramp set to be installed at the rear of the property.

The site is located near other properties owned by Mr Kumar, including 3–7 West Auckland Road, which houses Cockerton Pharmacy and an associated flat.
Planning proposals comply with the Darlington Local Plan according to the council.
These include sustainable development and district centre policies, and enhancing local health infrastructure.
Highways and Environmental Health raised no objections to the proposal.
The practice will not have any new parking spaces, but it is located 200m from Cockerton Car Park, which has sufficient public parking.
Nearby pedestrian crossings and bus stops will also enhance connectivity.
The local area is primarily commercial, so the practice is not expected to have a significant impact on residential or public amenity.
No objections or comments were received from the public during the notification period.
Conditions of approval include installation of a rear access ramp, adherence to approved plans, and noise controls on external equipment.
The development must commence within three years and must be carried out in accordance with the approved plans.
